If I replace a wired modem/router with a wireless one, will the computers on the existing wired network need to be re-configured? Or is it just a case of configuring the new modem/router to connect with my ISP.

The second half is correct. Once the new router is connected to the ISP, any computer plugged in will work. Wireless ones should use security (WPA).
